What are the main factors in setting the fan speed for a 3 speed Trane 5 ton indoor coil heat pump? Is the superheat checked after several settings are checked, or just once with lo speed????

Ideally you want to set the fan speed and then check your charge. The fan speed should be set to whatever the manufacturer suggests for a given capacity (350-450 CFM per Ton). To set the fan speed you need to measure the external static pressure and then use the blower performance data to help you determine what fan speed should be used. Once you have the correct airflow you can check and adjust the charge. Hope this helps...
